Mechanism of Action of Loxapine

Loxapine produce its antipsychotic activity by blocking the post synaptic receptor in the CNS.

Absorption: Loxapine is rapidly absorbed after oral administration. Distribution: Drug is distributed widely in the body in protein bound form. Metabolism: Loxapine is extensively metabolised in the liver. Excretion: Most of them excreted in urine and also in faeces.

Psychoses

Interactions for Loxapine

Alcohol & other CNS depressants: Additive CNS depression.
TCAs & Antiparkinsonism drugs: Anticholinergic effects potentiated.
Clonidine & Methyldopa: Hypotensive effect of these drugs inhibited.
MAOIs: Orthostatic hypotension with MAOIs.

Typical Dosage for Loxapine

Initial dose: 20mg / day in 2 divided doses. Increased dose is useful in severe schizophrenic patients.
Maintenance dose: 60 - 100 mg / day in 2 - 4 divided doses.
Maximum dose: 250 mg / day

Storage Requirements for Loxapine

Store at 15 - 30 degree C in a tightly closed container. Protect from heat and light. Keep out of the reach of children.

Effects of Missed Dosage of Loxapine

Take the missed dose as soon as noticed. If it is the time for next dose then take that dose and skip the missed dose. Continue the regular schedule. Do not double the dose.

Effects of Overdose of Loxapine

Give supportive measures and symptomatic treatment. Gastric lavage can be done to remove the drug from the body. Activated charcoal can be given to reduce the absorption of the drug. Treat hypotension with IV fluid, seizure with parenteral Diazepam and arrhythmia with Phenytoin.
