Latanoprost + Benzalkonium Chloride Pharmacology
Latanoprost + Benzalkonium Chloride
About Latanoprost + Benzalkonium ChlorideProstaglantin analoguesMechanism of Action of Latanoprost + Benzalkonium ChlorideLatanoprost is a prostaglandin F2a analogue. Specifically, Latanoprost is a prostanoid selective FP receptor agonist that is believed to reduce the intraocular pressure (IOP) by increasing the outflow of aqueous humor. Studies in animals and man suggest that the main mechanism of action is increased uveoscleral outflow. Elevated IOP represents a major risk factor for glaucomatous field loss. The higher the level of IOP, the greater the likelihood of optic nerve damage and visual field loss.Pharmacokinets of Latanoprost + Benzalkonium ChlorideAbsorption :Latanoprost is well absorbed through the cornea where the isopropyl ester prodrug is hydrolyzed to the acid form. Peak concentration is reached 2 hrs after topical administration.
Metabolism
Primarily hepatic (none except hydrolysis in the eye). Latanoprost is an isopropyl ester prodrug. It is hydrolyzed by esterases in the cornea to latanoprost acid, which is biologically active. The portion of the latanoprost acid that reaches the systemic circulation is metabolized primarily by the liver to 1,2-dinor and 1,2,3,4-tetranor metabolites by fatty acid beta-oxidation.

About Benzalkonium ChlorideA nitrogenous cationic surfactant, Quaternary ammonium compound, Antiseptic,Disinfectant, A vaginal contraceptive spermicidal agent.Mechanism of Action of Benzalkonium ChlorideThe mechanism of bactericidal/microbicidal action is due to disruption of intermolecular interactions. This cause dissociation of cellular membrane bilayers, which compromises cellular permeability controls and induces leakage of cellular contents. Other biomolecular complexes within the bacterial cell can also undergo dissociation. It also inactivates the enzyme produced by different microorganisms. It also has spermicidal action. They work by damaging and killing sperm in the vagina. Therefore, the sperm are not able to travel from the vagina into the uterus and fallopian tubes, where fertilization usually takes place.Pharmacokinets of Benzalkonium ChlorideN/AOnset of Action for Benzalkonium ChlorideN/ADuration of Action for Benzalkonium ChlorideN/AHalf Life of Benzalkonium ChlorideN/ASide Effects of Benzalkonium Chloride1.
