About Mitomycin
A methylazirinopyrroloindoledione derivative isolated from Streptomyces caespitosus or Streptomyces lavendulae, An anti-neoplastic antibiotic.
Mechanism of Action of Mitomycin
Mitomycin converted into bifunctional and trifunctional alkylating agent invivo by enzymatic or chemical reduction. These agents alkylate at the position of N6 of adenine and O6 and N7 position of guanine. In addition it breaks the single stranded DNA and chromosomes. It also inhibit the generation of super oxide and hydroxyl radicals by selectively act on hypoxide cells.

Absorption- Rapidly absorbed after intravenous administration
Distribution- Widely distributed in the body
Metabolism- Metabolized in the liver to its active and inactive metabolites. Active metabolites are deactivated in the kidney, spleen, brain and heart.
Excretion-Excreted through urine, bile and feces

Interactions for Mitomycin
Vinca alkaloids: Acute shortness of breath and severe bronchospasm have occured following the use of vinca alkaloids in patients who had previously or concurrently received mitomycin.
Typical Dosage for Mitomycin
Intravenous-
Adult-
10 to 20mg/metre square as a single dose every 4 to 8 weeks
2mg/metre square/day for 5 days. This cycle is repeated for 6 to 8 weeks

Effects of Missed Dosage of Mitomycin
