Mechanism of Action of Nabumetone
The parent compound is a prodrug, which undergoes hepatic biotransformation to the active component, 6-methoxy-2-naphthylacetic acid (6MNA), that is a potent inhibitor of prostaglandin synthesis, most likely through binding to the COX-1 and COX-2 receptors.Its action is more potent on COX2. This results its antipyretic, analgesics and anti-inflammatory action.

Absorption- Well absorbed after oral administration. Rapidly it undergoes biotransformation in the plasma to its active metabolite 6-methoxy-2-naphthylacetic acid (6MNA).
Distribution- Widely distributed in the body. 99% bound to plasma proteins
Metabolism-It is metabolized in the liver to its inactive metabolites. Excretion- Excreted through urine and feces
