A Beta-2 adrenergic agonist, A tocolytic(uterine relaxant).
Mechanism of Action of Ritodrine
Ritodrine is ?2 adrenergic agonist. It binds to ?2 adrenergic receptor on outer membrane of myometrial cell and activates adenylyl cyclase. This will result in an increase in cyclic adenosine monophosphate (cyclic AMP) level and decreases intracellular calcium concentration. The decrease of calcium concentration leads to a relaxation of uterine smooth muscle.

Absorption: Ritodrine is completely absorbed after intravenous administration.
Metabolism: It is metabolised in the liver by glucuronide conjugation.
Excretion: It is excreted mainly in the urine.

Typical Dosage for Ritodrine
Adult:
Oral: 10 mg 30 minutes before the termination of intravenous infusion, then 10 mg every 2 hours for 24 hours, then maintenance dose is 10 to 20 mg 4 - 6 times daily until the 37th week of gestation the dose depending on uterine activity and unwanted effects.
Maximum dose: 120mg / day
Injection: initial dose: 0.05mg/minute as IV infusion, dose is then increased by 0.05mg every 10 minutes until therapeutic effect is obtained, maximum dose: 0.35mg/minute

Effects of Missed Dosage of Ritodrine
Take the missed dose as soon as noticed and if it is the time for next dose then skip the missed dose. Continue the regular schedule. Do not double the dose.
Effects of Overdose of Ritodrine
Give supportive measures and symptomatic treatment. The absorption of oral overdose can be reduced by emptying stomach and by administration of activated charcoal. If needed ? blocker Propranolol can be used as antidote for Ritodrine overdose.
