Mechanism of Action of Tolfenamic Acid
Tolfenamic acid acts by inhibition of Prostaglandin (PGs) synthesis and their release at the site of injury. Prostaglandins cause tenderness and amplify the action of other algesics. Tolfenamic acid does not affect the tenderness induced by PGs but block the pain producing mechanism induced by inflammatory mediators by inhibiting the enzyme cyclo-oxygenase.

Absorption: Tolfenamic acid is readily absorbed from the gastrointestinal tract.
Distribution: It is distributed mainly in protein bound form. It is distributed into breast milk also. Metabolism: It is metabolised in the liver. Both metabolite and the drug are conjugated with glucuronic acid.
Excretion: 90% of the drug is excreted in the urine and the remaining in faeces.
