About SucralfatePepsin inhibitor, Anti-secretory, Gastrointestinal Agent, antiulcer.Mechanism of Action of SucralfateSucralfate is a complex of sucrose, sulfate and aluminium hydroxide. In the stomach (pH<4) Sucralfate undergoes polymerization and form viscous sticky polymers that adhere to the surface of ulcers and coat the ulcer.
It also protects the ulcer surface from further injury by inhibiting the enzyme pepsin which hydrolysis mucosal proteins. Sucralfate increases the production of Prostaglandins and protect the lining of the stomach. It also binds to the epithelial growth factor and Fibroblast factor and repair the mechanism of stomach lining.
Pharmacokinets of SucralfateAbsorption: Sucralfate is poorly absorbed after oral administration.
Distribution: It is mainly distributed to the ulcer site and the absorbed drug is distributed to body tissues.
Excretion: It is excreted mainly in the faeces and the absorbed drug is excreted in the urine.

Interactions for SucralfateAntibacterials (ciprofloxacin, norfloxacin, ofloxacin and tetracycline), Antiepileptics (phenytoin), Antifungals (ketoconazole), Cardiac glycosides (digoxin), Thyroxine: Sucralfate may reduce the absorption of these drugs.
Anticoagulants: Reduced efficacy of warfarin.
Antacids (aluminium containing) :Total body burden of aluminium may be increased.
Cimetidine, Ranitidine and Theophylline: Reduced bioavailability of these drugs.
Quinidine: Serum quinidine levels may be reduced.Typical Dosage for SucralfateAdult:
Oral dose: 4 g / day in 2 - 4 divided doses drug to be taken 1 hour before meals and at bed time.
Suspension: 10 ml to be taken 4 times daily.
Children: not recommended
