Dihydrofolate reductase inhibitor, Pyrimidine derivative, Anti-infective(bacteriostatic antibiotic).
Mechanism of Action of Trimethoprim
Trimethoprim is a bacteriostatic antibiotic and acts by interfering with the action of bacterial dihydrofolate reductase, inhibiting synthesis of tetrahydrofolic acid. Tetrahydrofolic acid is an essential precursor in the de novo synthesis of the DNA nucleosides thymidine and uridine. Bacteria are unable to take up folic acid from the environment and are thus dependent on their own de novo synthesis.

Mechanism of Action of Sulfadiazine
Sulfadiazine is a short-acting sulfonamide. Sulfadiazine is a structural analogs and competitive antagonists of para-aminobenzoic acid (PABA). They inhibit normal bacterial utilization of PABA for the synthesis of dihydrofolic acid, a compound that bacteria must be able to make in order to survive for the synthesis of folic acid, an important metabolite in DNA synthesis. It is usually given with Pyrimethamine in the treatment of Chloroquine resistant Plasmodium falciparum in a fixed dose formulation.
Sulfadiazine has also been used with other sulfonamides, particularly Sulfamerazine and Sulfadimidine, to reduce the problems of low solubility in urine.

Children:
Initial dose: 75 mg/kg, then 150 mg/kg daily in divided doses to a maximum of 6 g daily. Infants less than 2 months:
Congenital toxoplasmosis: 50 mg/kg twice daily for 12 months
Toxoplasmosis (Immuno compromised patients): 4 to 6 g daily in 4 divided doses for at least 6 weeks, followed by 2 to 4 g daily
For the prophylaxis of rheumatic fever: 500mg - 1 g daily
